Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. Genetic variations in the IL-12B gene, specifically the rs3212227 CC genotype, are linked to rheumatoid arthritis (RA) susceptibility. Elevated serum IL-12 levels in RA patients correlate with disease severity and hypertension.

Background:

  • Rheumatoid arthritis (RA) is an autoimmune disease influenced by genetic factors, particularly polymorphisms in cytokine genes.
  • Interleukin-12 (IL-12) is crucial for balancing T-helper 1 (Th1) and T-helper 2 (Th2) cell responses.

Purpose of the Study:

  • To investigate the association between IL-12B gene single nucleotide polymorphisms (SNPs) (rs3212227 and rs17860508) and rheumatoid arthritis (RA) susceptibility and severity.
  • To evaluate serum IL-12 levels in RA patients and their correlation with clinical manifestations.

Main Methods:

  • Case-control study involving 634 RA patients and 341 healthy controls.
  • Genotyping performed using PCR-RFLP and high-resolution melting analysis for IL-12B SNPs.
  • Serum IL-12 concentrations measured by ELISA.

Main Results:

  • The rs3212227 CC genotype of IL-12B was significantly more frequent in RA patients than in controls (P=0.037).
  • A trend towards higher frequency of the rs3212227 C allele was observed in RA patients (P=0.07).

  • The IL-12B rs3212227 polymorphism (CC genotype) may contribute to genetic susceptibility to RA in the Polish population.
  • Elevated serum IL-12 levels are associated with RA disease activity and comorbidities like hypertension.
