IP3/DAG Signaling Pathway01:11

IP3/DAG Signaling Pathway

15.4K
Membrane lipids such as phosphatidylinositol (PI) are precursors for several membrane-bound and soluble second messengers. Specific kinases phosphorylate PI and produce phosphorylated inositol phospholipids. One such inositol phospholipids are the  phosphatidylinositol-4,5 bisphosphate [PI(4,5)P2], present in the inner half of the lipid bilayer. Upon ligand binding, GPCR stimulates Gq proteins to turn on phospholipase Cꞵ. Activated phospholipase Cꞵ cleaves PI(4,5)P2 and...

Phosphoinositides and PIPs01:42

Phosphoinositides and PIPs

10.4K
Phosphoinositides are a group of phospholipids containing a glycerol backbone with two fatty acid chains and a phosphate attached to a myoinositol sugar ring. The inositol head group extends into the cytoplasm, where it is modified by adding phosphate groups to form phosphatidylinositol phosphates or PIPs.
Different phosphoinositides are synthesized and recruited on the cytosolic face of the plasma membrane. The localization of specific phosphoinositides concentrated in separate membrane...

Rab Cascades01:25

Rab Cascades

3.7K
Rab GTPases act in a regulated cascade during membrane fusion, helping the lipid bilayers mix. The Rab family of proteins are active when bound to GTP, and inactive when bound to GDP. Hence, they act as guanine nucleotide-dependent molecular switches. Rab-GTP recognizes and binds to long or short-range tethering proteins to capture the target vesicle. These tethers coordinate with SNAREs on the vesicle and the target membrane to assemble the trans SNARE complex that locks the mixing bilayers.

Rab Proteins01:14

Rab Proteins

5.3K
Rab proteins constitute the largest family of monomeric GTPases, of which 70 members are present in humans. Rab proteins and their effectors regulate consecutive stages of vesicle transport such as vesicle transport, docking, and fusion to the correct recipient membrane.
Rab proteins switch between a cytosolic, GDP-bound inactive state and a membrane-anchored, GTP-bound active state. By themselves, Rabs show slow rates of GDP/GTP exchange and GTP hydrolysis. Thus, Rab proteins are considered...

Assembly of Signaling Complexes01:30

Assembly of Signaling Complexes

6.9K
Multiprotein signaling complexes are formed in a dynamic process involving protein-protein interactions at the cytoplasmic domain of transmembrane receptors or enzymatic and non-enzymatic proteins associated with the receptor. These complexes ensure the activation and propagation of intracellular signals that regulate cell functions.
Interaction domains in cell signaling
Interaction domains recognize exposed features of their binding partners containing post-translationally modified sequences,...

Coat Assembly and GTPases01:33

Coat Assembly and GTPases

4.6K
Vesicles incorporate different coat protein subunits in different cell locations, which changes the properties of the coat, such as the shape and geometry of the transport vesicles. Thus, vesicle coat proteins also play a significant role in cargo selection.
Coat assembly depends on the local availability of phosphatidylinositol phosphates or PIPs and GTP-binding proteins. Adaptor proteins, which link the coat proteins to the membrane, bind to these PIPs and play a crucial role in controlling...

Area of Science:

  • Genetics
  • Molecular Biology
  • Epigenetics

Background:

  • Paternally Expressed Gene 3 (Peg3) is an imprinted gene encoding a zinc finger DNA-binding protein.
  • Peg3 resides within a cluster of Kruppel-Associated Box (KRAB) zinc finger genes.
  • Exon 7 of Peg3 shares sequence similarity with KRAB-A domains.

Purpose of the Study:

  • To investigate the functional classification of Peg3 within the KRAB zinc finger gene family.
  • To determine if Peg3 interacts with known KRAB-associated co-repressors.
  • To elucidate the mechanism by which Peg3 regulates downstream genes.

Main Methods:

  • Co-immunoprecipitation assays to detect protein-protein interactions.
  • Analysis of Peg3 exon 7 for functional domains.
  • Chromatin immunoprecipitation (ChIP) assays.
  • Promoter activity assays.

Main Results:

  • The PEG3 protein was successfully co-immunoprecipitated with KAP1, a known KRAB-interacting co-repressor.
  • Experiments confirmed that Peg3 exon 7 is essential for its physical interaction with KAP1.
  • ChIP and promoter assays indicated that PEG3 mediates gene repression via KAP1.

Conclusions:

  • Peg3 is identified as a KRAB-containing zinc finger protein.
  • Peg3 physically interacts with the co-repressor KAP1.
  • Peg3 likely regulates downstream gene expression through KAP1-mediated repression.
