Background:

  • Peripartum sleep disturbances are common in humans but lack adequate animal models.
  • Understanding physiological changes during pregnancy and postpartum is crucial for maternal health.

Purpose of the Study:

  • To establish a Wistar rat model for studying peripartum sleep-wakefulness disorders.
  • To investigate adaptive changes in anxiety levels associated with pregnancy and postpartum.

Main Methods:

  • Surgically implanted electroencephalogram (EEG) and electromyogram (EMG) electrodes in female rats for sleep-wakefulness (S-W) monitoring.
  • Recorded S-W patterns throughout pregnancy, postpartum, and after weaning.
  • Assessed anxiety levels using the elevated plus maze test.

Main Results:

  • Sleep increased during pregnancy, mainly light non-REM sleep during the dark phase.
  • Increased non-REM sleep delta power was observed postpartum, with fragmented sleep, particularly during the day.
  • Anxiety levels increased during the third trimester of pregnancy, decreasing after parturition.

Conclusions:

  • This study provides the first comprehensive report on diurnal/nocturnal variations in sleep, delta power, and anxiety across the peripartum period in an animal model.
  • The developed Wistar rat model is suitable for preclinical drug trials and research on peripartum sleep disorders.
