Long-term Depression01:03

Long-term Depression

3.5K
Long-term depression, or LTD, is one of the ways by which synaptic plasticity—changes in the strength of chemical synapses—can occur in the brain. LTD is the process of synaptic weakening that occurs over time between pre and postsynaptic neuronal connections. The synaptic weakening of LTD works in opposition to synaptic strengthening by long-term potentiation (LTP) and together are the main mechanisms that underlie learning and memory.

Drugs that Stabilize Microtubules01:15

Drugs that Stabilize Microtubules

2.8K
Microtubules are dynamic structures that undergo cycles of catastrophe and rescue. The microtubules play a central role in cell division by forming the spindle apparatus for segregating the chromosomes. This makes them ideal targets for regulating dividing cells in tumors and malignant cancer cells. Microtubule stabilizing drugs help stabilize the microtubule formation and promote its polymerization. Paclitaxel was the first microtubule stabilizing agent used as anticancer drug in chemotherapy...

Area of Science:

  • Neuroscience
  • Oncology
  • Pharmacology

Background:

  • Chemotherapeutic agents are vital for systemic cancer treatment.
  • Adverse side effects, including cognitive deficits, reduce survivors' quality of life.
  • Cognitive impairments affect memory, executive function, and processing speed, sometimes long-term.

Purpose of the Study:

  • To investigate the effects of paclitaxel on learning and memory in rats.
  • To examine cognitive challenges and rule-learning in paclitaxel-treated rats.

Main Methods:

  • Rats were administered paclitaxel, a taxane-derived chemotherapeutic agent.
  • A source memory task was used to assess spatial and episodic memory.
  • Cognitive challenges and rule-learning were evaluated under varying experimental contingencies.

Main Results:

  • Paclitaxel treatment did not impair spatial or episodic memory.
  • Paclitaxel-treated rats showed no increased susceptibility to cognitive challenges.
  • Despite intact memory, paclitaxel impaired the learning of new rules, indicating reduced sensitivity to changing contingencies.

Conclusions:

  • Paclitaxel treatment has specific cognitive effects, dissociating memory function from rule-learning abilities.
  • Chemotherapy-induced cognitive impairments may involve a nuanced impact on different cognitive domains.
  • Findings highlight the incongruent vulnerability of episodic memory and new learning following paclitaxel treatment.
