Functional single-nucleotide polymorphisms (SNPs) in innate immune genes like Toll-like receptors (TLRs) and mannose-binding lectin (MBL) may protect against severe chronic obstructive pulmonary disease (COPD). These genetic variations are linked to milder disease phenotypes and fewer exacerbations.
Area of Science:
- Immunogenetics
- Pulmonology
- Molecular Biology
Background:
- The innate immune system is crucial in chronic obstructive pulmonary disease (COPD) pathogenesis.
- Functional single-nucleotide polymorphisms (SNPs) in innate immune genes may influence COPD phenotype and prognosis.
Purpose of the Study:
- To investigate the role of Toll-like receptor 2 (TLR2) and Toll-like receptor 4 (TLR4) SNPs and mannose-binding lectin (MBL) deficiency in Greek COPD patients.
- To assess the combined effects of these genetic factors on COPD severity.
Main Methods:
- Genotyping of functional TLR2 and TLR4 SNPs (TLR2-R753Q, TLR4-D299G, TLR4-T399I).
- Assessment of genotypic mannose-binding lectin (MBL) deficiency.
- Analysis of genetic associations with COPD stage and exacerbation frequency in 114 Greek COPD patients.
Main Results:
- TLR4-D299G and TLR4-T399I SNPs were associated with an earlier stage of COPD (p=0.003 and p=0.009).
- Absence of analyzed polymorphisms, including TLR2-R753Q and MBL deficiency, correlated with more severe COPD phenotypes and increased exacerbations (p=0.045).
Conclusions:
- Presence of functional innate immune SNPs, such as TLR polymorphisms and MBL deficiency, may offer a protective effect on COPD phenotype.
- These findings suggest a potential role for immunogenetics in modulating COPD severity and outcomes.
