Summary
Genetic variations in the adiponectin (ADIPOQ) gene, specifically rs2241767 and the T-G-A haplotype, are linked to an increased risk of ischemic stroke (IS) and large-artery atherosclerosis (LAA) in the Chinese population.
Area of Science:
- Genetics
- Cardiovascular Disease
- Molecular Biology
Background:
- Adiponectin, an adipose tissue-specific protein, exhibits anti-inflammatory and antiatherogenic properties, suggesting a protective role against stroke.
- Genetic polymorphisms in the adiponectin (ADIPOQ) gene may influence susceptibility to ischemic stroke (IS).
Purpose of the Study:
- To investigate the association between three ADIPOQ gene polymorphisms (rs1501299, rs2241767, and rs3774261) and their haplotypes with IS and its subtypes in a Chinese population.
- To determine if specific ADIPOQ gene variants are risk factors for large-artery atherosclerosis (LAA) and small-vessel occlusion (SVO).
Main Methods:
- Genotyping of ADIPOQ gene polymorphisms rs1501299, rs2241767, and rs3774261 in 385 IS patients and 418 controls using polymerase chain reaction-restriction fragment length polymorphism analysis.
- Statistical analysis including univariate and multivariate logistic regression to assess the association between genotypes, haplotypes, and IS subtypes.
- Haplotype analysis was performed for the T(rs1501299)-G(rs2241767)-A(rs3774261) combination.
Main Results:
- The A allele frequency of rs2241767 was higher in IS and LAA patients compared to controls (P=.028 and P=.017).
- The GG genotype and dominant model (GG+AG) of rs2241767 were associated with an increased risk of LAA (P=.040 and P=.034).
- Multivariate analysis revealed that the GG genotype of rs2241767 was independently associated with IS and LAA (OR=1.822, P=.037; OR=2.051, P=.038, respectively).
- The T(rs1501299)-G(rs2241767)-A(rs3774261) haplotype showed a moderately increased risk for IS and LAA (OR=1.595, P=.025; OR=1.709, P=.031, respectively).
- No significant association was found for rs1501299 and rs3774261 polymorphisms with IS or its subtypes.
Conclusions:
- The ADIPOQ gene rs2241767 polymorphism is tentatively associated with susceptibility to ischemic stroke and large-artery atherosclerosis in the Chinese population.
- The T(rs1501299)-G(rs2241767)-A(rs3774261) haplotype may also contribute to the risk of ischemic stroke and large-artery atherosclerosis in this cohort.

Single Nucleotide Polymorphisms-SNPs
19.3K
A single nucleotide polymorphism or SNP is a single nucleotide variation at a specific genomic position in a large population. It is the most prevalent type of sequence variation found in the human genome. Point mutations that occur in more than 1% of the population qualify as SNPs. These are present once every 1000 nucleotides on an average in the human genome. Replacement of a purine with another purine (A/G) or a pyrimidine with another pyrimidine (C/T) is known as a transition. In contrast,...

Pharmacogenetics of Drug Targets: β₂-Adrenergic Receptors, Apo E, Thymidylate Synthase
60
Genetic polymorphisms in drug targets have emerged as critical determinants of interindividual variability in drug response and toxicity. Pharmacogenomic investigations increasingly focus on identifying these variations to personalize and optimize therapeutic interventions. A drug target may be a receptor, enzyme, or signaling protein involved in pharmacologic responses or disease-related pathways. While early pharmacogenetic studies focused primarily on drug metabolism, current research...

Advances in genomics have profoundly influenced drug discovery by increasing both the speed and accuracy of pharmaceutical development. Pharmacogenomics, which examines how genetic variation influences drug response, facilitates the identification of novel therapeutic targets and enables patient stratification for personalized treatment. These strategies contribute to improved drug efficacy, minimized adverse effects, and more efficient clinical trial design.Mapping genetic differences...

Pharmacogenetic Phenotypes: Alterations in Pharmacokinetics, Drug Targets and Biologic Milieu
81
Genetic variations significantly influence drug response through pharmacokinetics, receptor interactions, and biologic milieu modifications. Pharmacokinetic alterations impact drug metabolism and clearance, affecting efficacy and toxicity. Variants in drug-metabolizing enzymes, such as CYP2C9 and CYP2C19, alter drug activation and elimination. For example, CYP2C9 loss-of-function variants require lower warfarin doses to prevent excessive bleeding, while CYP2C19 variants reduce clopidogrel...

Principles of Pharmacogenetics: Types of Genetic Variants
69
The human genome is over 99.9% identical between individuals, yet genetic differences exist at millions of bases. The human genome contains approximately 3 million variant positions per individual, many of which are heterozygous, contributing to genetic diversity and individual traits. Genetic variations include single-nucleotide polymorphisms (SNPs), insertions, deletions, and copy number variations (CNVs).SNPs, the most common variation, involve single-base changes in DNA. These can be...
