Area of Science:

  • Biochemistry
  • Biotechnology
  • Endocrinology

Background:

  • Plasma leptin, secreted by adipose tissue, is crucial in physiological and pathological processes.
  • Altered leptin levels are associated with metabolic disorders like obesity and diabetes.

Purpose of the Study:

  • To develop and evaluate a protein biochip-based sandwich assay for detecting plasma leptin.
  • To compare the biochip assay's performance against the established ELISA method.

Main Methods:

  • A protein biochip assay was utilized to detect plasma leptin in 96 participants (45 healthy, 28 obese, 23 diabetic).
  • Assay sensitivity was determined, and results were compared with those from a standard ELISA.

Main Results:

  • Plasma leptin levels were significantly higher in obese and diabetic patients compared to healthy individuals (p < 0.001).
  • The biochip assay demonstrated a low limit of detection (0.006 μg/mL) and good correlation with ELISA (R² = 0.948).

Conclusions:

  • A protein biochip approach provides a viable method for plasma leptin detection.
  • This biochip assay shows potential as a substitute for conventional ELISA methods in clinical diagnostics.
