Summary
This study identified novel long noncoding RNA (lncRNA) candidates involved in abdominal aortic aneurysm (AAA) pathogenesis. One specific lncRNA, lnc-ARG, was found to decrease 5-lipoxygenase (ALOX5) expression and reactive oxygen species production.
Area of Science:
- Molecular Biology
- Genomics
- Cardiovascular Research
Background:
- Abdominal aortic aneurysm (AAA) is a life-threatening condition with complex pathogenesis.
- Long noncoding RNAs (lncRNAs) are increasingly recognized for their roles in various diseases, including cardiovascular conditions.
Purpose of the Study:
- To investigate the role of lncRNAs in the pathogenesis of abdominal aortic aneurysm (AAA).
- To identify novel lncRNA candidates associated with AAA development.
Main Methods:
- Microarray analysis of lncRNA and mRNA expression in AAA and normal aortic tissues.
- Real-time quantitative reverse-transcription PCR (qRT-PCR) for expression verification.
- Computational analysis to predict lncRNA-mRNA targeting relationships and protein-coding potential.
- In vitro experiments in HeLa cells to assess the effect of lnc-ARG on ALOX5 expression and reactive oxygen species (ROS) production.
Main Results:
- Identification of 3,688 differentially expressed lncRNAs and 3,007 differentially expressed mRNAs between AAA and normal tissues.
- Discovery of 1,284 differentially expressed long intergenic noncoding RNAs and 206 differentially expressed enhancer-like lncRNAs.
- Construction of a co-expression network linking differentially expressed lncRNAs and mRNAs.
- Demonstration that lnc-ARG, located upstream of ALOX5, reduces ALOX5 mRNA levels and ROS production in HeLa cells.
Conclusions:
- This study highlights the significant involvement of lncRNAs in AAA pathogenesis.
- Novel lncRNA candidates, including lnc-ARG, have been identified as potentially crucial players in AAA development and progression.
