Summary

Circular RNAs (circRNAs) accumulate in aging mouse brains but not hearts. This suggests specific roles for these stable RNA molecules in the aging nervous system.

Background:

  • Circular RNAs (circRNAs) are stable RNA molecules formed by back-splicing.
  • CircRNAs are abundant in neural tissues, hinting at potential roles in brain function.

Purpose of the Study:

  • To investigate circRNA accumulation during aging in mouse brain and heart tissues.
  • To determine if age-related changes in circRNAs are specific to neural tissues.

Main Methods:

  • Total RNA sequencing (RNA-seq) of young and aged mouse cortex, hippocampus, and heart.
  • Validation of circRNA expression using RT-qPCR and Northern blot analysis.

Main Results:

  • 6,791 distinct circRNAs were detected, including 675 novel ones.
  • Significant upregulation of circRNAs was observed in aged neural tissues (cortex and hippocampus).
  • No global trend of circRNA upregulation was found in aged heart tissues.
  • Age-accumulation of brain circRNAs was largely independent of host gene expression.

Conclusions:

  • CircRNA accumulation is a prominent feature of aging in the mouse nervous system.
  • These findings suggest that circRNAs may have specific biological functions related to neural aging.
