Researchers sequenced a novel virus, Nesidiocoris tenuis virus 1, from the plant bug Nesidiocoris tenuis. This RNA virus has unique genomic features and infects over 28% of sampled insects.
Area of Science:
- Virology
- Genomics
- Entomology
Background:
- Nesidiocoris tenuis is an important insect pest in agriculture.
- The presence of novel viruses can impact insect populations and agricultural practices.
Purpose of the Study:
- To determine the complete genome sequence of a novel virus found in Nesidiocoris tenuis.
- To characterize the genomic structure and phylogenetic relationships of the newly identified virus.
- To investigate the prevalence of this virus in field populations of N. tenuis.
Main Methods:
- RNA sequencing (RNA-seq) was employed to obtain the viral genome sequence.
- Rapid amplification of cDNA ends (RACE) was used for full-length genome determination.
- Phylogenetic analysis was conducted using deduced amino acid sequences of viral proteins.
Main Results:
- The complete genome of N. tenuis virus 1, a novel single-stranded RNA virus, was sequenced (3970 nucleotides).
- The genome contains two open reading frames (ORFs) encoding RNA helicase and RNA-dependent RNA polymerase domains.
- Phylogenetic analysis suggests N. tenuis virus 1 is a prototype of a new unclassified virus type, related to Blackford virus.
- Prevalence varied by location, with 28.32% of sampled N. tenuis testing positive.
Conclusions:
- N. tenuis virus 1 represents a novel viral species with unique genomic characteristics.
- The virus infects a significant proportion of field populations of N. tenuis.
- Further research is needed to understand the virus's impact on its host and potential role in biological control.
