Tumor necrosis factor-alpha (TNF-α) enhances human dental pulp stem cell (hDPSC) migration by upregulating integrin α6. Suppressing integrin α6 reduces TNF-α-induced hDPSC chemotaxis, revealing a novel cytokine-adhesion receptor interaction.
Area of Science:
- Stem cell biology
- Immunology
- Regenerative medicine
Background:
- Stem cell migration is crucial for tissue repair and inflammatory responses.
- Tumor necrosis factor-alpha (TNF-α) is upregulated in inflamed tissues.
- Human dental pulp stem cells (hDPSCs) express integrin α6, linked to cell migration.
Purpose of the Study:
- To investigate the effect of TNF-α on hDPSC migration.
- To explore the relationship between TNF-α and integrin α6 expression in hDPSCs during migration.
Main Methods:
- Established hDPSC cultures and determined α6 integrin subunit expression.
- Evaluated TNF-α's effect on hDPSC migration using transwell and scratch assays.
- Assessed TNF-α-mediated migration with and without integrin α6, using siRNA to suppress integrin α6 expression.
Main Results:
- TNF-α significantly increased hDPSC migration.
- TNF-α upregulated integrin α6 expression.
- Knockdown of integrin α6 reduced TNF-α-directed hDPSC chemotaxis.
Conclusions:
- TNF-α activation of integrin α6 increases hDPSC migratory activity.
- A novel association between TNF-α and the integrin α6 chain regulates hDPSC migration.
