Background:

  • Glycoprotein non-metastatic melanoma protein B (GPNMB) is a type I transmembrane protein found in various cell types, including hematopoietic lineages.
  • Previous studies identified GPNMB as highly expressed during early and intermediate eosinophil development.

Purpose of the Study:

  • To investigate the expression pattern and functional role of GPNMB during eosinophil differentiation from cord blood (CB) CD34+ hematopoietic stem cells.
  • To determine if GPNMB influences cell proliferation during eosinophil development.

  • Utilized a 24-day in vitro culture system for CB CD34+ cells differentiating into eosinophils.
  • Employed Western blot and confocal microscopy to analyze GPNMB expression levels.
