Area of Science:

  • Forensic Science
  • Genetics
  • Biometrics

Background:

  • Lip prints are unique biometric identifiers with potential for personal identification.
  • Limited research exists on establishing lip prints as legally admissible biological evidence.
  • The study aimed to explore DNA extraction and short tandem repeat (STR) loci profiling from lip prints.

Purpose of the Study:

  • To determine the feasibility of isolating DNA from lip prints on diverse surfaces.
  • To obtain a comprehensive short tandem repeat (STR) loci profile from lip print impressions.
  • To assess the potential of lip prints as a source of biological evidence in forensic investigations.

Main Methods:

  • Procurement of 12 lip prints on surfaces like tissue paper, cotton cloth, ceramic tile, and glass.
  • Development of latent lip prints using fingerprint powder and collection of lipstick-coated prints.
  • DNA isolation, quantification, and amplification using Identifiler™ kit for 15 STR loci typing.

Main Results:

  • Sufficient DNA quantities were extracted from all tested lip print impressions.
  • Successful typing of 15 STR loci was achieved in seven samples.
  • Thirteen to fourteen STR loci were successfully typed in the remaining samples.

Conclusions:

  • Lip prints can serve as valuable biological evidence at crime scenes.
  • DNA extraction and STR loci typing from lip prints validate their forensic significance.
  • Genetic profiles from lip prints on items like cups and napkins are useful for suspect identification.
