Do mesothelin/MUC16 interactions facilitate adenocarcinoma metastases to intracranial meningiomas?
1Department of Pathology and Laboratory Medicine, University of Rochester Medical Center, Rochester, New York, India.
Background:
Meningiomas have been shown to express mesothelin, a high affinity binding site for MUC16, a transmembrane protein on adenocarcinoma cells. The mechanisms underlying adenocarcinoma metastases to meningiomas may provide insight into tumor-to-tumor metastases and adenocarcinoma metastases to leptomeningeal cells.
Methods:
Two meningiomas containing metastases from adenocarcinomas were identified and evaluated immunohistochemically for the expression and localization of mesothelin and MUC16.
Results:
Both meningiomas show extensive mesothelin immunoreactivity, and the adenocarcinomas metastatic to the meningiomas show mesothelin and MUC16 immunoreactivity at the interface with meningioma.
Conclusions:
Interactions between MUC16 and/or mesothelin on the cell membrane of adenocarcinoma cells with mesothelin on meningioma cells may facilitate adenocarcinoma metastases to meningiomas and possibly the leptomeninges.
Insights
Adenocarcinomas can metastasize to meningiomas by interacting with mesothelin (a protein on meningiomas) and MUC16 (a protein on adenocarcinoma cells). This interaction may also drive cancer spread to the brain's lining.
Area of Science:
- Oncology
- Cell Biology
- Pathology
Background:
- Meningiomas express mesothelin, a binding site for MUC16 found on adenocarcinoma cells.
- Understanding adenocarcinoma metastasis to meningiomas offers insights into tumor-to-tumor spread and leptomeningeal carcinomatosis.
Purpose of the Study:
- To investigate the role of mesothelin and MUC16 in adenocarcinoma metastasis to meningiomas.
- To evaluate the expression and localization of mesothelin and MUC16 in tumor interfaces.
Main Methods:
- Immunohistochemical evaluation of two meningioma samples with adenocarcinoma metastases.
- Assessing mesothelin and MUC16 expression and cellular localization.
Main Results:
- Both meningiomas exhibited significant mesothelin immunoreactivity.
- Metastatic adenocarcinomas showed mesothelin and MUC16 expression at the tumor interface.
Conclusions:
- Cellular interactions between MUC16/mesothelin on adenocarcinomas and mesothelin on meningiomas may facilitate metastasis.
- This mechanism could be relevant for adenocarcinoma spread to meningiomas and the leptomeninges.
