Summary

Rheumatoid arthritis (RA) can cause periodontal inflammation. This study found that intracellular NAMPT in periodontal ligament cells drives RA-related gum disease by regulating inflammatory genes.

Background:

Purpose of the Study:

  • To verify if RA mediates periodontitis-like phenotypes in experimental RA mouse models.
  • To explore the role of nicotinamide phosphoribosyltransferase (NAMPT) in periodontal inflammation during RA.

Main Methods:

  • Utilized collagen-induced arthritis (CIA) and TNF-α transgenic (TG) mouse models.
  • Examined NAMPT expression in periodontal ligament (PDL) tissues and human PDL cells.
  • Investigated the effects of NAMPT overexpression and inhibition (FK866, nicotinamide) on PDL cells and in CIA mice.

Main Results:

  • NAMPT expression was upregulated in RA mouse PDL tissues and human PDL cells stimulated by IL-1β and TNF-α.
  • NAMPT overexpression increased pro-inflammatory cytokines, chemokines, and matrix-degrading enzymes in PDL cells.
  • Inhibition of intracellular NAMPT (iNAMPT) suppressed RA-induced periodontal inflammation and tissue damage in mouse models.

Conclusions:

  • RA is associated with periodontal inflammation and bone loss in experimental models.
  • Intracellular NAMPT in PDL cells plays a key role in RA-mediated periodontal inflammation.
  • iNAMPT regulates catabolic genes (IL6, IL8, CCL5, COX-2, MMP1, MMP3) involved in RA-associated periodontitis.
