Purpose of the Study:

  • To investigate the relationship between self-reported executive functioning (using the Behavior Rating Inventory of Executive Function - BRIEF-A) and genetic polymorphisms in dopaminergic and serotoninergic systems.
  • To examine the impact of neuropsychological and personality traits on these gene-executive function associations in a non-clinical population.

Main Methods:

  • One hundred healthy adults completed the BRIEF-A, personality inventories (SPQ-74, STAI, MMPI), and executive function neuropsychological tests.
  • Genotyping was performed for polymorphisms in DRD4, COMT, DRD2, HTR2A, and SLC6A4 genes.
  • Statistical analyses explored main effects, genotype effects, and correlations.

Main Results:

  • A significant association was found between the SLC6A4 5-HTTLPR polymorphism and BRIEF-A scores.
  • Specific BRIEF-A scales (Plan/Organize, Task Monitor) and the Metacognition index were significantly affected by genotype.
  • Individuals with the short allele of 5-HTTLPR reported fewer executive functioning problems compared to long allele homozygotes. This effect persisted even when controlling for trait anxiety, which was a strong predictor of BRIEF-A scores.

Conclusions:

  • The SLC6A4 5-HTTLPR polymorphism may play a role in self-perceived everyday executive functioning, particularly in planning and monitoring.
  • While personality traits like anxiety strongly predict self-reported executive function, the genetic influence of 5-HTTLPR remains significant.
  • This study highlights the importance of considering genetic factors in understanding individual differences in everyday executive functioning.
