The Retinoblastoma Gene01:20

The Retinoblastoma Gene

4.9K
Tumor suppressor genes are normal genes that can slow down cell division, repair DNA mistakes, or program the cells for apoptosis in case of irreparable damage. Hence, they play an essential role in preventing the proliferation of damaged cells.
The first-ever tumor suppressor gene called Rb was identified in retinoblastoma - a rare eye tumor in children. In inherited forms of the disease, a child inherits one defective copy of the Rb gene, which predisposes them to retinoblastoma. However,...

Abnormal Proliferation02:23

Abnormal Proliferation

5.3K
Under normal conditions, most adult cells remain in a non-proliferative state unless stimulated by internal or external factors to replace lost cells. Abnormal cell proliferation is a condition in which the cell's growth exceeds and is uncoordinated with normal cells. In such situations, cell division persists in the same excessive manner even after cessation of the stimuli, leading to persistent tumors. The tumor arises from the damaged cells that replicate to pass the damage to the...

mTOR Signaling and Cancer Progression03:03

mTOR Signaling and Cancer Progression

5.0K
The mammalian target of rapamycin or mTOR protein was discovered in 1994 due to its direct interaction with rapamycin. The protein gets its name from a yeast homolog called TOR. The mTOR protein complex in mammalian cells plays a major role in balancing anabolic processes such as the synthesis of proteins, lipids, and nucleotides and catabolic processes, such as autophagy in response to environmental cues, such as availability of nutrients and growth factors.
The mTOR pathway or the...

Summary

Monocyte Chemoattractant protein-induced protein 1 (MCPIP1) is downregulated in clear cell renal cell carcinoma (ccRCC). Reduced MCPIP1 levels promote ccRCC development by increasing pro-cancer factors.

Area of Science:

  • Molecular Biology
  • Oncology
  • Biochemistry

Background:

  • Monocyte Chemoattractant protein-induced protein 1 (MCPIP1), also known as Regnase-1, regulates inflammatory responses and transcript stability.
  • MCPIP1 plays a role in controlling transcription factors like NF-κB and AP1.
  • ZC3H12a is the gene encoding MCPIP1.

Purpose of the Study:

  • To investigate the role of MCPIP1 in clear cell renal cell carcinoma (ccRCC) development.
  • To analyze the regulation and function of MCPIP1 in cancer progression.

Main Methods:

  • Analysis of MCPIP1 transcript and protein levels in ccRCC patient samples versus normal tissues.
  • Utilized Caki-1 cells as a model system to study MCPIP1 expression and function.
  • Investigated the impact of hypoxia, HIF2α, and proteasome activity on MCPIP1 levels.
  • Assessed the effect of MCPIP1 on HIF1α, HIF2α, VEGFA, GLUT1, IL-6, mTOR, and Akt.

Main Results:

  • MCPIP1 levels are significantly downregulated in ccRCC tissues compared to normal tissues.
  • Hypoxia and HIF2α were identified as key factors reducing MCPIP1 expression, which is dependent on proteasome activity.
  • MCPIP1 negatively regulates HIF1α and HIF2α, impacting HIF2α transcript stability.
  • Enhanced MCPIP1 expression led to decreased VEGFA, GLUT1, and IL-6 transcripts and reduced mTOR and Akt activity.

Conclusions:

  • MCPIP1 downregulation is implicated in ccRCC development.
  • MCPIP1 acts as a tumor suppressor by inhibiting key pro-cancer pathways involving HIFs, VEGFA, GLUT1, IL-6, mTOR, and Akt.
