Summary

Toxoplasma gondii infection causes eye disease. This study found that Interleukin-10 (IL-10) produced by T cells may protect against active toxoplasmic retinochoroiditis, suggesting a potential therapeutic target.

Purpose of the Study:

  • To investigate cytokine production by T cells in patients with active toxoplasmic retinochoroiditis (TR).
  • To compare cytokine profiles between active TR patients and healthy controls.
  • To explore the role of specific cytokines, like IL-10, in the disease's pathology.

Main Methods:

  • Studied 18 patients with active TR and 15 healthy controls (IgG+ and negative).
  • Incubated peripheral blood mononuclear cells with T. gondii antigen (STAg).
  • Analyzed cytokine expression (TNF, IL-10, IFN-γ) on CD4+ and CD8+ T cells using flow cytometry.

Main Results:

  • Baseline cytokine expression was higher in TR patients compared to controls.
  • In vitro stimulation with STAg increased cytokine production in TR patients' lymphocytes.
  • T cells were identified as the primary source of IL-10, IFN-γ, and TNF.
  • A negative correlation was found between ocular lesion size and IL-10 expression by CD4+ T cells.

Conclusions:

  • T cells are the main producers of IL-10 in patients with active toxoplasmic retinochoroiditis.
  • IL-10 expression appears to play a protective role in active TR.
  • These findings highlight the immunomodulatory role of IL-10 in ocular toxoplasmosis.
