Nivolumab shows promise for metastatic anal cancer. This immunotherapy was well-tolerated and effective in a phase 2 trial for squamous cell carcinoma of the anal canal (SCCA).

Background:

  • Squamous cell carcinoma of the anal canal (SCCA) is a rare cancer linked to human papillomavirus (HPV).
  • There is no established treatment for metastatic SCCA.
  • HPV oncoproteins in SCCA can increase immune checkpoint proteins like PD-1, helping cancer evade the immune system.

Purpose of the Study:

  • To evaluate the effectiveness and safety of nivolumab, an anti-PD-1 antibody, in patients with metastatic SCCA.
  • To determine the response rate of nivolumab in treatment-refractory metastatic SCCA patients.

Main Methods:

  • A single-arm, multicenter, phase 2 clinical trial was conducted.
  • 37 patients with treatment-refractory metastatic SCCA received nivolumab (3 mg/kg) every two weeks.
  • The primary endpoint was response rate using RECIST v1.1 criteria.
