Area of Science:

  • Biotechnology
  • Immunology
  • Molecular Biology

Background:

  • Phage display is a key technology for generating specific monoclonal antibodies.
  • Traditional antibody generation methods have limitations.
  • Presenting membrane antigens requires specific techniques.

Purpose of the Study:

  • To describe a whole cell panning procedure for antibody selection.
  • To utilize a Fab-based antibody library for this process.
  • To incorporate primary cell-based screening for enhanced selectivity.

Main Methods:

  • Implementing whole cell panning with a Fab-based antibody library.
  • Utilizing membrane-embedded target antigens in their native conformation.
  • Employing primary cell-based screening for selectivity assessment.

Main Results:

  • Demonstration of a functional whole cell panning protocol.
  • Successful selection of specific antibodies against membrane antigens.
  • Validation of primary cell screening for improved antibody specificity.

Conclusions:

  • Whole cell panning is a powerful approach for antibody discovery.
  • This method preserves antigen conformation for better antibody selection.
  • The described procedure enhances the specificity of selected antibodies.
