Reduced antioxidant capacity and superoxide dismutase (SOD) levels are observed in X-linked adrenoleukodystrophy (ALD) patients. Plasma SOD may predict cerebral ALD (cALD) onset, enabling earlier therapeutic intervention.
Area of Science:
- Biochemistry
- Genetics
- Neurology
Background:
- X-linked adrenoleukodystrophy (ALD) can progress to a fatal cerebral form (cALD).
- The underlying mechanisms driving this phenotypic switch are not fully understood.
- Identifying predictive biomarkers is crucial for timely intervention.
Purpose of the Study:
- To investigate differences in antioxidant capacity and superoxide dismutase (SOD) activity between ALD phenotypes (cALD, adrenomyeloneuropathy [AMN]), female carriers, and healthy controls.
- To assess the potential of these markers as predictors for cALD development.
Main Methods:
- Collected blood plasma and monocyte samples from healthy controls, female carriers, and male patients with AMN or cALD.
- Assayed total antioxidant capacity and SOD activity using commercially available kits.
- Correlated antioxidant levels with clinical data, including MRI severity scores.
Main Results:
- Significantly reduced antioxidant capacity and SOD activity were observed across ALD phenotypes compared to controls.
- Plasma SOD activity showed a strong inverse correlation with cALD brain MRI severity.
- Longitudinal data indicated decreasing SOD activity preceding cerebral diagnosis.
Conclusions:
- Plasma SOD levels are significantly diminished in patients with ALD, particularly in the cerebral form.
- Plasma SOD activity may serve as a valuable predictive biomarker for cerebral ALD.
- Early identification of cALD risk through SOD monitoring could facilitate prompt, life-saving therapies.
