Abstract

Insights

Colchicine effectively reduces kidney tissue injury and fibrosis in a rat model of ureteral obstruction. Higher doses demonstrated more significant anti-fibrotic and anti-apoptotic effects, suggesting a dose-dependent therapeutic benefit.

Area of Science:

  • Nephrology
  • Pharmacology
  • Cell Biology

Background:

  • Colchicine stabilizes microtubules and may reduce tissue injury.
  • The dose-dependent effects of colchicine on kidney injury were previously unknown.
  • Unilateral ureteral obstruction (UUO) is a model for studying kidney fibrotic disease.

Purpose of the Study:

  • To investigate the anti-fibrotic and apoptotic effects of varying colchicine doses.
  • To determine if colchicine's renoprotective effects are dose-dependent in a UUO model.

Main Methods:

  • Thirty-six Sprague-Dawley rats were used in a UUO model.
  • Rats were divided into sham and UUO groups, treated with vehicle or colchicine (30, 60, 100 μg/kg/day) for 7 days.
  • Kidney tissue was analyzed for markers of fibrosis, apoptosis, and tubulointerstitial damage.

Main Results:

  • Colchicine dose-dependently reduced acetylated α-tubulin and TGF-β immunoreactivity in obstructed kidneys.
  • Higher colchicine doses significantly attenuated tubulointerstitial damage and apoptosis.
  • Expression of cleaved caspase-3, ED-1, and fibronectin decreased with colchicine treatment.

Conclusions:

  • Appropriate colchicine dosage exhibits anti-fibrotic and anti-apoptotic effects in obstructed kidneys.
  • Optimal colchicine dosing is crucial for maximizing the prevention of renal disease progression.
  • Further evaluation is needed to determine optimal clinical doses for preventing kidney disease.

Drugs that Destabilize Microtubules

Microtubules are dynamic structures and can be regulated by microtubule targeting agents (MTAs). Microtubule destabilizing drugs are a class of MTAs that destabilize and prevent microtubules' polymerization. Both natural and synthetic chemicals can be found under this class of drugs. Vincristine and vinblastine, two vinca alkaloids, and colchicine were among the first to be discovered. These drugs can affect cells in various ways, either by inducing a change in cell morphology, preventing...

Acute Kidney Injury IV: Diagnostic Studies and Prevention

Accurate diagnosis and effective prevention are critical in managing Acute Kidney Injury (AKI), which is linked to high mortality rates ranging from 10% to 80%. Timely recognition of at-risk patients and careful monitoring can significantly reduce the likelihood of kidney damage.Diagnostic Assessments:The diagnostic process starts with a comprehensive medical history to identify prerenal, intrarenal, and postrenal causes.Prerenal causes, such as dehydration, hypotension, or blood loss, should...

Drugs for Treatment of Ulcerative Colitis in IBD

Ulcerative colitis is a chronic inflammatory condition primarily affecting the colon and rectum. The primary drugs used in the treatment of ulcerative colitis are aminosalicylates. They exhibit anti-inflammatory and immunosuppressive properties. They modulate inflammatory mediators and inhibit the activity of nuclear factor κB (NF-κB). Aminosalicylates also reduce inflammation by inhibiting prostaglandin and leukotriene production and decreasing neutrophil chemotaxis and superoxide...

Acute Kidney Injury II: Pathophysiology

Acute kidney injury (AKI) causes are categorized into three primary categories based on the location of the injury: prerenal, intrarenal (or intrinsic), and postrenal causes. This classification guides clinical management and illustrates how different pathways can impair kidney function.Etiology and Pathophysiology of Acute Kidney Injury1. Prerenal causesEtiology: Prerenal Acute Kidney Injury, the most common type, occurs when reduced blood flow to the kidneys decreases filtration capacity...

Renal Corpuscle

The glomerulus and Bowman's capsule are two essential components of the nephron, which is the functional unit of the kidney. These microscopic structures play a critical role in the process of blood filtration to produce urine.
Glomerulus: Structure and Function
The glomerulus is a tiny, intricate network of capillaries located at the beginning of the nephron. It's enveloped by the Bowman's capsule and receives its blood supply from an afferent arteriole, which divides into numerous...

Factors Affecting Renal Clearance: Renal Impairment

Renal dysfunction significantly impairs the renal clearance of drugs, leading to potential complications in drug therapy. Renal failure, which can be caused by various factors, poses a significant challenge in the elimination of drugs from the body.
One condition associated with renal failure is uremia. Uremia is characterized by impaired glomerular filtration and fluid accumulation in the body. This condition hinders the renal clearance of drugs, resulting in drug accumulation and potential...
