Purpose of the Study:

  • To adapt and validate frailty indices for aged Fischer (F344) rats.
  • To assess the predictive value of rat frailty for survival outcomes.

Main Methods:

  • Male Fischer (F344) rats (17 months old) underwent behavioral testing.
  • Tests included forelimb wire suspension (strength), rotarod (speed), open field (activity), and inclined screen (endurance).
  • Frailty was defined by poor performance in 2 (mildly frail) or 3-4 (frail) tasks.

Main Results:

  • 17.29% of rats were classified as mildly frail, and 2.26% as frail.
  • Mildly frail rats had a 3.8-fold increased mortality risk over 100 days.
  • Frail rats exhibited a 27.5-fold increased mortality risk over 100 days.

Conclusions:

  • The developed frailty index effectively identifies frail aged Fischer (F344) rats.
  • This index predicts increased mortality risk in this aging rat model.
  • The findings support the use of this tool for preclinical frailty research.
