Summary

Human blood platelets activate sodium-hydrogen exchange (Na+/H+) when their cytoplasm acidifies. This process helps regulate platelet pH, similar to mechanisms in other cells.

Area of Science:

  • Biochemistry
  • Cell Biology
  • Physiology

Background:

  • Cytoplasmic acidification triggers Na+/H+ exchange in human blood platelets.
  • This exchange results in intracellular alkalinization, measurable with fluorescent pH indicators.

Purpose of the Study:

  • To investigate the properties and role of the Na+/H+ exchanger in human platelets.
  • To characterize the kinetic and inhibitory profiles of platelet Na+/H+ exchange.

Main Methods:

  • Intracellular pH monitoring using fluorescent indicators.
  • Platelet volume analysis with a Coulter Counter in Na-propionate medium.
  • Assessment of inhibitor sensitivity (amiloride derivatives) and ion specificity (Na+, Li+, K+, Rb+, Cs+).

Main Results:

  • Platelet Na+/H+ exchange activity was confirmed and characterized.
  • The exchanger demonstrated specificity for Na+ and Li+, with a Km for Na+ of 75-90 mM.
  • Inhibitor studies showed sensitivity to amiloride and its derivatives, with competitive inhibition observed.

Conclusions:

  • The Na+/H+ exchanger in human platelets shares properties with exchangers in other cell types.
  • This exchanger plays a significant role in regulating intracellular pH within platelets.
