Background:

  • Platelet alpha 2-adrenergic receptor function is often studied as a potential biomarker in psychiatric disorders.
  • Previous research suggests alterations in adrenergic receptor function in schizophrenia and depression.

Purpose of the Study:

  • To investigate alpha 2-adrenergic receptor sensitivity in platelets of patients with schizophrenia and depression.
  • To compare platelet cyclic adenosine-3',5'-monophosphate (cAMP) responses to norepinephrine (NE) and prostaglandin E1 (PGE1) between patient groups and controls.

Main Methods:

  • Blood samples were collected from drug-free patients with schizophrenia, patients with depression, and healthy controls.
  • Platelet aggregation and cAMP levels were measured in response to NE and PGE1 stimulation.
  • The inhibitory effect of NE on the PGE1-stimulated cAMP response was quantified.

Main Results:

  • The absolute inhibition of the cAMP response to PGE1 by NE was smaller in schizophrenic and depressive patients compared to controls.
  • This difference was attributed to a lower baseline cAMP response to PGE1 in the patient groups.
  • No significant differences in NE effects on cAMP production were observed between actively ill and remitted patients.

Conclusions:

  • Platelet alpha 2-receptor sensitivity, assessed via NE's effect on cAMP production, does not appear to be altered in schizophrenia or depression.
  • The observed differences in NE inhibition are likely due to alterations in baseline cAMP signaling rather than alpha 2-receptor function itself.
