Summary

The rd8 mouse model shows reduced negative scotopic threshold responses (nSTRs) and smaller a- and c-waves, indicating inner retinal dysfunction despite outer retinal effects.

Background:

  • The rd8 mouse is a model for retinal degeneration.
  • Previous studies suggested a late-onset electroretinographic (ERG) phenotype in rd8 mice.

Purpose of the Study:

  • To evaluate the electroretinographic (ERG) phenotype of rd8 mice.
  • To assess both conventional ERG and scotopic threshold responses (STRs) in rd8 mice.

Main Methods:

  • Tested 4-month-old wild-type (WT) and rd8 mice.
  • Recorded mass retinal responses (ERGs) using various visual stimuli.
  • Accumulated scotopic threshold responses (STRs) with dim flashes.

Main Results:

  • rd8 mice exhibited lower negative STR (nSTR) amplitudes compared to WT.
  • A- and c-wave amplitudes were smaller in rd8 mice under higher intensity stimuli.
  • B-wave amplitudes showed a non-significant trend towards larger values in rd8 mice.

Conclusions:

  • The significant reduction in nSTRs in rd8 mice suggests inner retinal dysfunction.
  • This finding is unexpected as rd8 mutation effects are primarily in the outer retina.
  • Further investigation may reveal novel insights into retinal cell biology and circuitry.
