Area of Science:

  • Neuroscience
  • Microscopy
  • Biotechnology

Background:

  • Understanding brain function necessitates high-resolution, 3D imaging of neural connections.
  • Current methods involving serial sectioning are labor-intensive and introduce artifacts.
  • Optical clearing techniques render tissues transparent for advanced imaging.

Purpose of the Study:

  • To review popular optical clearing techniques and their mechanisms.
  • To compare the advantages and disadvantages of various clearing approaches.
  • To present light-sheet microscopy principles and applications in neuroscience.

Main Methods:

  • Review of optical clearing methods (e.g., CLARITY, iDISCO).
  • Explanation of light-sheet fluorescence microscopy (LSFM) principles.
  • Analysis of applications in whole-brain imaging and connectomics.

Main Results:

  • Optical clearing enables deep tissue penetration and uniform transparency.
  • Light-sheet microscopy provides rapid, high-resolution 3D imaging of cleared specimens.
  • Combined techniques overcome limitations of traditional histological methods.

Conclusions:

  • Optical clearing coupled with light-sheet microscopy revolutionizes whole-brain imaging.
  • These advanced techniques facilitate comprehensive mapping of neural circuits.
  • Applications extend beyond neuroscience to other biological fields.
