T follicular helper (Tfh) cells are diverse. PD1-low Tfh-like cells enter follicles without B cell interaction and may precede CD4 memory cells, unlike PD1-high Tfh cells.
Area of Science:
- Immunology
- Cell Biology
Background:
- T follicular helper (Tfh) cells are crucial for adaptive immunity, driving germinal center (GC) formation.
- Tfh populations are heterogeneous, with PD1-high Tfh cells known to be important in GC responses.
- The precise function and developmental pathway of PD1-low Tfh-like cells remain largely unknown.
Purpose of the Study:
- To investigate the function and differentiation pathway of PD1-low Tfh-like cells.
- To compare the requirements for PD1-low Tfh-like cell entry into follicles versus PD1-high GC-Tfh cells.
- To elucidate the role of B cell interactions and CXCR5 in Tfh subset differentiation.
Main Methods:
- Flow cytometry to identify and quantify Tfh subsets (PD1-high and PD1-low).
- Studies on B cell dependency and follicular homing mechanisms.
- Analysis of Tfh differentiation pathways involving B cell interactions and CXCR5.
Main Results:
- PD1-low Tfh-like cells, similar to PD1-high Tfh cells, depend on B cells for their development.
- Entry of PD1-low Tfh-like cells into follicles is independent of CXCR5 and cognate B cell interactions.
- Differentiation into PD1-high Tfh cells requires MHC class II-B cell interactions and CXCR5, suggesting a stepwise Tfh differentiation pathway.
Conclusions:
- A novel Tfh differentiation pathway is proposed, starting B cell-independent, progressing through non-cognate B cell interactions, and culminating in cognate interactions for GC-Tfh formation.
- PD1-low Tfh-like cells exhibit early cytokine responses.
- PD1-low Tfh-like cells may serve as precursors for CD4 memory T cells, highlighting their distinct functional role.

Diversity of Antigen Receptors
Before encountering any antigen, lymphocytes express these receptors. On B cells, the antigen receptor is a membrane-bound antibody molecule called BCR; on T cells, it is a T cell receptor or TCR. B and T cell receptors are composed of two...
