Purpose of the Study:

  • To evaluate casein-loaded DCs for their ability to induce Th1 immune responses.
  • To assess the potential of casein-loaded DCs as a therapeutic approach for allergic conditions.
  • To investigate immune cell therapy using DCs in the context of cow's milk allergy.

Main Methods:

  • Immature bone marrow-derived DCs were loaded with casein (protein or mRNA) or GFP mRNA.
  • DC markers and proinflammatory cytokine production were analyzed.
  • Lymphocyte proliferation and cytokine secretion (IFN-γ, IL-10, TGF-β, IL-4) were measured.

Main Results:

  • Casein-loaded DCs exhibited increased DC marker expression compared to immature DCs.
  • TNF-α and IL-12 levels increased, while IL-6 decreased in casein-DC treatments.
  • Casein-loaded DCs stimulated lymphocyte proliferation and IFN-γ, IL-10, and TGF-β production, but not IL-4.

Conclusions:

  • Casein-loaded DCs partially elicit Th1 immune responses.
  • This approach shows promise for immune cell therapy in managing cow's milk allergy.
  • Further research into DC-based AIT for allergies is warranted.
