Low expression of plant homeodomain finger 2 (PHF2) correlates with aggressive clear cell renal cell carcinoma (CCRCC). This suggests PHF2 may serve as a prognostic biomarker for CCRCC patients.
Area of Science:
- Oncology
- Epigenetics
- Molecular Biology
Background:
- Clear cell renal cell carcinoma (CCRCC) is linked to adipogenic differentiation.
- Histone modification, particularly by histone demethylase plant homeodomain finger 2 (PHF2), plays a role in adipogenesis.
- PHF2 may function as a tumor suppressor through epigenetic regulation of p53 and its reduced expression is observed in other cancers.
Purpose of the Study:
- To investigate the expression of PHF2 in CCRCC specimens.
- To determine the correlation between PHF2 expression and clinicopathological features and prognosis of CCRCC.
Main Methods:
- Immunohistochemistry was used to examine PHF2 expression.
- The study included 254 CCRCC tissues and 56 non-neoplastic renal tissues.
- Tissue microarray blocks were prepared for analysis.
Main Results:
- High PHF2 expression was observed in 59.1% of CCRCC cases, while 40.1% showed low expression.
- High PHF2 expression was significantly associated with lower Fuhrman nuclear grade, smaller tumor size, and lower overall stage.
- High PHF2 expression correlated with longer cancer-specific survival and progression-free survival.
Conclusions:
- Low PHF2 expression is associated with increased aggressiveness and poorer prognosis in CCRCC.
- PHF2 expression levels may serve as a potential prognostic biomarker for CCRCC.

The Retinoblastoma Gene
The first-ever tumor suppressor gene called Rb was identified in retinoblastoma - a rare eye tumor in children. In inherited forms of the disease, a child inherits one defective copy of the Rb gene, which predisposes them to retinoblastoma. However,...
