    Summary

    ACC-9447, a topical beta adrenergic antagonist, showed a significant 24-hour intraocular pressure reduction in normal subjects. However, this effect was not statistically different from placebo in this initial study.

    Background:

    • Topical ocular medications often cause systemic side effects.
    • ACC-9447 is a novel beta adrenergic antagonist designed to minimize systemic absorption.
    • It utilizes blood esterases for rapid deactivation post-absorption.

    Purpose of the Study:

    • To evaluate the intraocular pressure (IOP) response to ACC-9447 in normal human subjects.
    • To assess the efficacy of ACC-9447 across a range of concentrations (0.2% to 4.0%).
    • To compare the IOP-lowering effect of ACC-9447 against a placebo control.

    Main Methods:

    • A randomized, fellow-eye controlled study involving 17 normal human subjects.
    • Participants received topical ACC-9447 in one eye and placebo in the fellow eye.
    • IOP was measured nine times over 24 hours, with repeat testing at increasing concentrations over six days.

    Main Results:

    • Eyes treated with ACC-9447 demonstrated a statistically significant reduction in IOP from baseline over 24 hours.
