Developmental changes in the rabbit sinus node action potential and its response to adrenergic agonists.

K W Hewett, M R Rosen

The Journal of Pharmacology and Experimental Therapeutics
    |November 1, 1985

    Summary

Young rabbits show developmental changes in sinus node response to beta-adrenergic stimulation, with shorter sinus cycle lengths. Alpha-adrenergic stimulation had no age-dependent effect on sinus node function in developing or adult rabbits.

    Area of Science:

    • Cardiology
    • Developmental Biology
    • Pharmacology

    Background:

    • The sinoatrial node (SAN) is the primary pacemaker of the heart.
    • Understanding developmental changes in cardiac electrophysiology is crucial for pediatric cardiology.
    • Adrenergic receptor signaling plays a key role in regulating heart rate.

    Purpose of the Study:

    • To investigate age-related changes in rabbit sinus node transmembrane potentials.
    • To examine the effects of beta-adrenergic and alpha-adrenergic stimulation on sinus node function during development.

    Main Methods:

    • Standard microelectrode techniques were employed.
    • Experiments were conducted on sinus node cells from 1-10 day old and adult rabbits.
    • Sinus node cells were superfused with Tyrode's solution at 37°C and exposed to adrenergic agonists.

    Related Experiment Videos

    Main Results:

    • Maximum diastolic potential remained unchanged with age.
    • Action potential amplitude was lower in young rabbits (days 1-3) compared to adults.
    • Young sinus nodes exhibited shorter sinus cycle lengths (SCL) and responded to lower isoproterenol concentrations for SCL reduction.
    • Alpha-adrenergic stimulation with phenylephrine showed no age-dependent effects on SCL or action potential characteristics.
    • The inhibitory effect of alpha-adrenergic stimulation, observed in other cardiac tissues, was absent in the sinus node, likely due to its membrane potential range.

    Conclusions:

    • Developmental changes in the sinus node's response to beta-adrenergic stimulation are evident.
    • The sinus node does not exhibit age-dependent changes in response to alpha-adrenergic stimulation.
    • Voltage-dependency of alpha-adrenergic effects explains its absence in the sinus node.
