    Summary

    Cigarette smoking significantly impedes asbestos fiber clearance in guinea pigs. Smokers retained more asbestos fibers, particularly shorter ones, increasing potential lung disease risk.

    Background:

    • Asbestos exposure is linked to serious lung diseases.
    • The role of smoking in asbestos clearance is not fully understood.

    Purpose of the Study:

    • To investigate the effect of cigarette smoking on the clearance of inhaled asbestos fibers in a guinea pig model.

    Main Methods:

    • Guinea pigs received amosite asbestos via intratracheal instillation.
    • Animals were divided into nonsmokers and two smoking groups (pre- and post-exposure).
    • Asbestos fiber concentration and size were analyzed at 1 week and 1 month.

    Main Results:

    • Nonsmokers showed a 6-fold decrease in asbestos burden from 1 week to 1 month.
    • Smoking groups exhibited no significant decrease in asbestos burden over the same period.
    • Mean fiber length increased in nonsmokers but decreased in smokers; shorter fibers were retained more.

    Conclusions:

    • Cigarette smoking impairs asbestos fiber clearance, primarily by retaining short fibers.
    • This increased pulmonary asbestos burden in smokers may contribute to higher disease rates in asbestos-exposed workers who smoke.
