Background:

  • Platelet (PLT) concentrate storage variables show donor-dependent differences.
  • Previous retrospective studies indicated a donor influence on PLT quality.

Purpose of the Study:

  • To prospectively investigate the donor effect on platelet concentrate (PC) quality.
  • To identify donor characteristics associated with superior or inferior PC storage performance.

Main Methods:

  • Male donors were selected based on Day 8 PC pH values (good >7.0, poor <6.7).
  • Donors completed health and lifestyle questionnaires.
  • PCs were stored for 12 days and analyzed for in vitro quality, including lactate production, CD62P expression, phosphatidylserine exposure, and mitochondrial membrane potential.

Main Results:

  • Poor donors (4/6) reported hypertension, high cholesterol/fat, or medication use, unlike good donors (0/6).
  • Good PCs exhibited lower lactate production and higher pH from Day 5 onwards.
  • Good PCs showed improved markers of activation and mitochondrial function at storage end, with only minor differences in functional properties.

Conclusions:

  • Platelet storage performance is predictable when donors are categorized as 'good' or 'poor' storers.
  • Metabolic variations may stem from differences in mitochondrial functionality.
  • Further research is required to elucidate underlying causes and implications for donors and blood products.
