This study profiles long non-coding RNA (lncRNA) expression in germinal center B-cell-like diffuse large B-cell lymphoma (GCB-DLBCL). Differentially expressed lncRNAs were identified, offering insights into GCB-DLBCL pathogenesis.
Area of Science:
- Molecular Biology
- Genomics
- Oncology
Background:
- Long non-coding RNAs (lncRNAs) are key regulators of cellular activities.
- The role of lncRNAs in germinal center B-cell-like diffuse large B-cell lymphoma (GCB-DLBCL) pathogenesis is largely unexplored.
Purpose of the Study:
- To investigate lncRNA expression profiles in GCB-DLBCL.
- To identify differentially expressed lncRNAs and predict their functional roles in GCB-DLBCL.
Main Methods:
- Microarray analysis was used to profile lncRNA expression in GCB-DLBCL cell lines and normal B lymphocytes.
- Quantitative reverse transcription polymerase chain reaction (qRT-PCR) validated lncRNA expression.
- Bioinformatic analyses, including Gene Ontology, pathway, and network analysis, were performed.
Main Results:
- A total of 21,539 lncRNAs were expressed across all samples.
- 1,648 lncRNAs were significantly upregulated and 2,671 were significantly downregulated in GCB-DLBCL cell lines (P<0.05).
- Pathway analysis implicated 64 upregulated and 62 downregulated pathways (P<0.05); a lncRNA-mRNA network was constructed.
Conclusions:
- This study identifies differentially expressed lncRNAs in GCB-DLBCL.
- The findings provide a foundation for understanding the functional roles of lncRNAs in GCB-DLBCL development.
- These results contribute to a deeper comprehension of GCB-DLBCL pathogenesis.
