Summary

Sucralfate can reduce superficial gastric lesions in patients taking non-steroidal anti-inflammatory drugs (NSAIDs). However, it does not significantly impact symptoms or ulcer formation when used with NSAIDs.

  • NSAID use is associated with significant gastrointestinal (GI) side effects, including superficial lesions and ulcers.
  • Sucralfate is a cytoprotective agent used to treat and prevent peptic ulcers.

Purpose of the Study:

  • To evaluate the efficacy of sucralfate in preventing NSAID-induced gastrointestinal side effects.
  • To assess the impact of sucralfate on GI symptoms, superficial lesions, and ulcer formation in patients on chronic NSAID therapy.

Main Methods:

  • A prospective, randomized, single-blind study was conducted from 1989 to 1992.
  • 176 patients requiring NSAIDs for over three months were recruited.
  • Patients received either placebo or sucralfate alongside NSAID therapy, with clinical and endoscopic assessments at baseline and after 6-8 weeks.

Main Results:

  • Gastrointestinal symptoms were reported by 30.6% of the placebo group (A) and 26.4% of the sucralfate group (B).
  • Superficial gastric lesions were observed in 36.5% of group A and 18.7% of group B.
  • Endoscopic ulcers were found in 2.4% of group A and 1.1% of group B.

Conclusions:

  • Simultaneous administration of sucralfate with chronic NSAID therapy reduces the incidence of superficial gastric lesions.
  • Sucralfate did not demonstrate a significant effect on alleviating GI symptoms or preventing ulcer formation in this patient group.
