    Summary

    This study questions the existence of a proton-pumping ATPase in plant vacuoles. New methods suggest that previously observed ATPase activity may not be directly linked to metabolite accumulation.

    Background:

    Purpose of the Study:

    • To investigate the validity of the proton-pumping ATPase hypothesis in plant vacuoles.
    • To re-evaluate the mechanisms of metabolite accumulation in plant vacuoles.

    • Utilized reverse-phase high-performance liquid chromatography (RP-HPLC) for simultaneous separation of adenine nucleotides.
    • Analyzed tonoplast vesicles isolated from Catharanthus roseus.
    • Observed H+-quenching of quinacrine fluorescence during ATP hydrolysis by vacuoles and tonoplast vesicles.
