    Growth hormone-releasing factor (GRF) neurons in the rat hypothalamus were mapped using immunohistochemistry. GRF fibers in the median eminence primarily originate from arcuate nucleus neurons, suggesting inhibitory regulation.

    Purpose of the Study:

    • To immunohistochemically characterize the growth hormone-releasing factor (GRF)-containing neuronal system in the rat hypothalamus.
    • To identify the origin and pathways of GRF fibers in the median eminence.
    • To investigate the influence of deafferentation and neonatal MSG treatment on GRF neurons.

    Main Methods:

    • Immunohistochemistry was used to visualize GRF and tyrosine hydroxylase (TH) immunoreactivity.
    • Colchicine was administered intraventricularly to enhance neuronal labeling.
    • Neonatal monosodium glutamate (MSG) treatment and hemidecortication were employed to study specific neuronal populations and their connections.

    Main Results:

    • GRF neurons were identified in the arcuate nucleus (group 1) and near the ventromedial nucleus (group 2).

    • Neonatal MSG treatment reduced group 2 neurons and median eminence fibers.

    • Median eminence GRF fibers predominantly originate from arcuate nucleus (group 1) neurons.
    • These neurons may be subject to inhibitory control from extraypothelamic regions.
    • GRF from group 2 neurons might function as a neurotransmitter-like substance.
