    Background:

    • The Human Leukocyte Antigen (HLA)-C locus plays a critical role in immune responses.
    • Understanding genetic polymorphism at the HLA-C locus is essential for transplantation and disease association studies.
    • Previous studies had identified some restriction fragment length polymorphisms (RFLPs) at this locus.

    Purpose of the Study:

    • To investigate further restriction fragment length polymorphism (RFLP) at the HLA-C locus using the pC250 DNA probe.
    • To identify new polymorphic sites and map their locations within HLA-C genes.
    • To characterize RFLPs associated with serologically defined Cw specificities and Cw blanks.

    Main Methods:

    • Genomic Southern blot hybridization using the HLA-C-specific DNA probe pC250.
    • Analysis of DNA from homozygous typing cells (Cw1-Cw8) and Cw blank samples.
    • Digestion with various restriction nucleases including Taq I, Pvu II, Bst XI, Nde I, Nci I, and Eco RI.
