Metformin effectively inhibits gastric cancer growth by targeting cancer stem cells (CSCs). This drug reduces tumor proliferation and self-renewal, offering a promising therapeutic strategy for this deadly disease.
Area of Science:
- Oncology
- Pharmacology
- Cancer Stem Cell Biology
Background:
- Gastric cancer is a leading cause of cancer mortality with a poor prognosis.
- Targeting cancer stem cells (CSCs) presents a novel therapeutic avenue for gastric cancer.
- Metformin is an antidiabetic drug with potential anticancer properties.
Purpose of the Study:
- To evaluate the anti-tumoural effects of metformin on gastric cancer.
- To determine if metformin targets gastric CSCs in vitro and in vivo.
- To assess metformin's impact on CSC proliferation, self-renewal, and differentiation markers.
Main Methods:
- In vitro studies using 2D and 3D cell cultures (tumorsphere assays) with gastric cancer cell lines and patient-derived primary tumor xenografts (PDXs).
- In vivo studies using mouse xenograft models.
- Analysis of cell cycle, CSC markers (CD44, Sox2), and differentiation markers (Kruppel-like factor 4, MUC5AC).
Main Results:
- Metformin induced cell cycle arrest, decreasing proliferation in 2D cultures.
- Metformin reduced tumorsphere formation in 3D cultures, indicating CSC targeting.
- Metformin altered CSC and differentiation marker expression and delayed tumor growth in vivo, reducing CSC self-renewal.
Conclusions:
- Metformin demonstrates significant anti-tumoural effects against gastric cancer.
- Metformin effectively targets gastric cancer stem cells, inhibiting their proliferation and self-renewal.
- Metformin represents a potential therapeutic strategy for gastric cancer by targeting CSCs.
