Summary

Tonsil-derived mesenchymal stem cell conditioned medium (T-MSC CM) reduces visceral fat and increases adiponectin. This T-MSC CM may offer therapeutic potential for obesity by enhancing adiponectin multimerization and reducing oxidative stress.

Area of Science:

  • Regenerative Medicine
  • Cell Biology
  • Metabolic Disease Research

Background:

  • Mesenchymal stem cells (MSCs) are crucial for regenerative medicine.
  • Tonsil-derived MSC conditioned medium (T-MSC CM) has previously shown visceral fat reduction.
  • Reduced visceral fat correlates with increased adiponectin levels.

Purpose of the Study:

  • To investigate the effect of T-MSC CM on adiponectin production.
  • To explore the impact of T-MSC CM on adiponectin secretion and multimerization.
  • To determine if T-MSC CM influences oxidative stress markers in adipocytes.

Main Methods:

  • T-MSC CM was administered to senescence-accelerated mice prone 6.
  • Adiponectin levels and expression were measured using RT-qPCR, Western blot, and ELISA.
  • 3T3-L1 adipocytes were treated with T-MSC CM and subjected to oxidative stress induction.

Main Results:

  • T-MSC CM injection reduced mouse weight and epididymal adipose tissue mass.
  • Significant increases in adiponectin expression and circulating levels (total and HMW) were observed.
  • T-MSC CM enhanced adiponectin secretion and multimerization in 3T3-L1 cells and reduced oxidative stress markers.

Conclusions:

  • T-MSC CM effectively reduces visceral fat and increases adiponectin levels in vivo.
  • T-MSC CM promotes adiponectin secretion and multimerization, potentially via ameliorating oxidative stress.
  • T-MSC CM demonstrates therapeutic potential for obesity and related metabolic disorders.
