Area of Science:

  • Pharmacognosy
  • Analytical Chemistry
  • Herbal Medicine Quality Control

Background:

  • Berberidis Cortex is a traditional herbal medicine with significant therapeutic applications.
  • Establishing robust quality control standards is crucial for ensuring the safety and efficacy of herbal products.
  • Existing quality control methods for Berberidis Cortex require refinement to meet modern analytical demands.

Purpose of the Study:

  • To establish quality standards for Berberidis Cortex.
  • To improve the quality control level of Berberidis Cortex through validated analytical methods.
  • To assess the suitability of Thin Layer Chromatography (TLC) and High-Performance Liquid Chromatography (HPLC) for quality assessment.

Main Methods:

  • Determination of water content, total ash, acid-insoluble ash, and alcohol-soluble extract as per Chinese Pharmacopoeia (2010 edition).
  • Qualitative identification using Thin Layer Chromatography (TLC).
  • Quantitative analysis of key alkaloids (magnoflorine, jatrorrhizine, palmatine, berberine) using High-Performance Liquid Chromatography (HPLC).

Main Results:

  • TLC identification demonstrated good resolution and clear spot separation.
  • Physicochemical parameters ranged as follows: water content (8.39%-12.23%), total ash (4.50%-9.96%), acid-insoluble ash (0.10%-0.69%), and alcohol-soluble extract (20.62%-37.13%).
  • Average alkaloid content: magnoflorine (5.98%), jatrorrhizine (0.63%), palmatine (0.30%), and berberine (2.50%).

Conclusions:

  • The developed TLC and HPLC methods are accurate and specific for Berberidis Cortex.
  • These validated methods provide a reliable basis for the future quality control of Berberidis Cortex.
  • The established quality standards contribute to ensuring the consistent quality and therapeutic value of Berberidis Cortex.
