Area of Science:

  • Pharmacology
  • Drug Metabolism
  • Clinical Pharmacology

Background:

  • Theophylline is a bronchodilator with a narrow therapeutic index.
  • Understanding drug interactions is crucial for safe theophylline use.
  • Temelastine is an antihistamine whose pharmacokinetic interactions require investigation.

Purpose of the Study:

  • To evaluate the effect of oral temelastine on the intravenous pharmacokinetics of theophylline.
  • To determine if temelastine alters the absorption, distribution, metabolism, or excretion of theophylline.

Main Methods:

  • A balanced, double-blind, crossover study was conducted in 10 healthy volunteers.
  • Subjects received oral temelastine (100 mg) or placebo twice daily for 7 days.
  • Theophylline (intravenous aminophylline 250 mg) pharmacokinetics were assessed.

Related Experiment Videos

Main Results:

  • No significant differences were observed in the volume of distribution, elimination rate constants, or elimination half-lives of theophylline.
  • Areas under the plasma concentration-time curve for theophylline were not significantly affected by temelastine administration.

Conclusions:

  • Temelastine does not significantly impact the pharmacokinetics of theophylline in healthy individuals.
  • Co-administration of temelastine with theophylline is unlikely to require dosage adjustments based on pharmacokinetic changes.
