Combined Effects of Drugs: Synergism

7.0K
Synergism is a useful mechanism where combining two or more drugs is more effective than each constituent used alone. Such combinations are also called supra-additive interactions. The drugs collectively enhance the final therapeutic effect by acting on different targets. Another advantage is that the low dose of each constituent drug is sufficient to achieve the desired effect. This helps reduce the duration of therapy and lower the adverse effects of these drugs.

Lipid-Lowering Drugs: Statins and Miscellaneous Agents

Hyperlipidemia, a medical condition often referred to as high cholesterol, is characterized by abnormally elevated levels of lipids in the bloodstream. When present in excess, these lipids, specifically cholesterol and triglycerides, can lead to serious health complications, often involving cardiovascular diseases. Illnesses like atherosclerosis, heart attacks, and pancreatitis have all been linked to untreated hyperlipidemia. This means controlling and regulating cholesterol and triglyceride...

Area of Science:

  • Pharmacognosy
  • Phytochemistry
  • Analytical Chemistry

Background:

  • Milk thistle (Silybum marianum) extract, silymarin, possesses antioxidant, anti-inflammatory, immunomodulatory, and hepatoprotective properties.
  • Research predominantly focuses on silybin, neglecting other flavonolignans and the neglected oil fraction, leading to reproducibility issues due to variable silymarin composition.
  • Variability in silymarin composition arises from processing, plant chemo-variety, and environmental factors, impacting scientific results.

Purpose of the Study:

  • To review and summarize High-Performance Liquid Chromatography (HPLC) separation techniques for determining flavonolignan content in silymarin.
  • To highlight the need for detailed comparative analysis of flavonolignan profiles in various silymarin preparations.
  • To emphasize the importance of the neglected oil fraction of milk thistle, containing fatty acids, sterols, tocopherol, and phospholipids.

Main Methods:

  • Review of existing literature on analytical separation and quantification of silymarin components.
  • Summary of various HPLC techniques employed for silymarin analysis.
  • Discussion of factors influencing silymarin composition and the potential applications of its neglected components.

Main Results:

  • A comprehensive overview of HPLC methods for silymarin analysis is presented.
  • The review underscores the lack of detailed comparative studies on flavonolignan profiles across different silymarin preparations.
  • The significance of the oil fraction, beyond its culinary uses, for industrial applications like biodiesel and polymer production is noted.

Conclusions:

  • Standardized analytical methods and comparative studies are crucial for improving the reproducibility of silymarin research.
  • Further investigation into the complete composition of silymarin, including the oil fraction, is warranted.
  • Milk thistle holds potential for diverse applications beyond its medicinal properties, including industrial uses.
