Background:

Purpose of the Study:

  • To investigate the distribution and nature of coated membrane structures involved in osteoclast endocytosis.
  • To differentiate between endocytic pathways in different osteoclast regions.

Main Methods:

  • Transmission electron microscopy (TEM) of osteoclasts from young rats.
  • Immunocytochemistry using anti-clathrin antibodies.
  • Examination of cells around developing tooth germs.

Main Results:

  • Coated pits and vesicles were absent in the ruffled border and clear zone of osteoclasts.
  • Clathrin-coated pits and vesicles were identified on the dorsal and lateral surfaces of actively resorbing osteoclasts.
  • Previously described coated membrane structures in the ruffled border are not associated with coated pits or vesicles.

Conclusions:

  • Osteoclast endocytosis involves distinct pathways: clathrin-mediated endocytosis for nutrient/macromolecule uptake on lateral/dorsal surfaces.
  • Bone mineral endocytosis at the ruffled border appears to be mediated by a non-clathrin-associated coated membrane structure.
