Summary

Stem cell factor (SCF) is elevated in overactive bladder (OAB) patients and promotes OAB symptoms. This suggests SCF may be a therapeutic target for OAB treatment.

Background:

  • Overactive bladder (OAB) is a condition affecting bladder function.
  • The role of specific molecular mediators in OAB pathogenesis is not fully understood.

Purpose of the Study:

  • To investigate the effect of stem cell factor (SCF) on overactive bladder (OAB) biological behavior.
  • To explore SCF's role as a potential mediator in inducing OAB.
  • To assess urinary SCF levels as a diagnostic biomarker for OAB.

Main Methods:

  • Immunohistochemistry to determine SCF localization in bladder tissue.
  • Measurement of urinary SCF levels in OAB patients and controls.
  • Administration of SCF to guinea pigs to assess urodynamic effects via cystometry.

Main Results:

  • SCF expression was found in the urothelium and throughout the bladder wall in guinea pigs and humans.
  • Urinary SCF and SCF/creatinine levels were significantly higher in OAB patients compared to controls.
  • SCF administration dose-dependently reduced inter-contraction interval and increased non-voiding contractions.

Conclusions:

  • SCF is present in the urinary bladder and its levels are elevated in OAB.
  • SCF may mediate OAB by binding to c-kit receptors on ICC-like cells, influencing bladder function.
