Simple spectrophotometry and spectrofluorimetry methods accurately estimate succinylcholine chloride (SUC) in pharmaceuticals. These assays utilize eosin Y for ion pair complexation or fluorescence quenching, offering sensitive quantification.
Area of Science:
- Analytical Chemistry
- Pharmaceutical Analysis
Background:
- Succinylcholine chloride (SUC) is a critical neuromuscular blocking agent.
- Accurate quantification of SUC in pharmaceutical preparations is essential for quality control.
Purpose of the Study:
- To develop simple, sensitive analytical methods for SUC estimation.
- To validate these methods for pharmaceutical preparations.
Main Methods:
- Spectrophotometry: Ion pair complex formation with eosin Y, measured at 550 nm.
- Spectrofluorimetry: Quenching of eosin Y native fluorescence by SUC, measured at 550 nm (excitation 480 nm).
- Both methods employed Britton Robinson buffer at pH 3.
Main Results:
- Spectrophotometry linearity: 0.5–15 μg/ml SUC.
- Spectrofluorimetry linearity: 0.05–1 μg/ml SUC.
- Successful application for succinylcholine ampoule estimation.
Conclusions:
- Developed spectrophotometric and spectrofluorimetric methods are simple, sensitive, and suitable for SUC quantification.
- The methods provide reliable results for pharmaceutical quality control.
- Reaction pathway elucidation supports the analytical basis.
