Summary

Programmed death-ligand 1 (PD-L1) is aberrantly expressed in most angiosarcomas, a rare and aggressive cancer. This finding is crucial for stratifying patients for immune checkpoint inhibitor therapies.

Area of Science:

  • Oncology
  • Immunology
  • Pathology

Background:

  • Angiosarcomas are rare, aggressive endothelial tumors with poor prognosis.
  • The PD-1/PD-L1 pathway is vital for tumor immune evasion.
  • PD-L1 expression is a potential biomarker for immunotherapy, but standardized IHC protocols are lacking.

Purpose of the Study:

  • To analyze the immunohistochemical (IHC) expression of PD-L1 in angiosarcoma tissues.
  • To establish reliable PD-L1 staining protocols for angiosarcomas.
  • To explore the potential of PD-L1 as a biomarker in angiosarcoma for immunotherapy stratification.

Main Methods:

  • Analysis of IHC expression of PD-L1 in a series of angiosarcoma samples.
  • Evaluation of PD-L1 staining across different body locations.
  • Correlation of PD-L1 expression with patient prognosis.

Main Results:

  • Aberrant PD-L1 expression was observed in approximately 66% of angiosarcoma samples.
  • PD-L1 expression showed no significant correlation with prognosis.
  • A reliable method for defining PD-L1 staining in angiosarcoma was established.

Conclusions:

  • PD-L1 is frequently expressed in angiosarcomas, supporting its potential role in immune evasion.
  • Standardized IHC assessment of PD-L1 is feasible in angiosarcoma.
  • This study provides a basis for stratifying angiosarcoma patients for PD-1/PD-L1 blockade immunotherapy.
