Area of Science:

  • Clinical Chemistry
  • Hematology
  • Point-of-Care Diagnostics

Background:

  • Accurate hemoglobin estimation is crucial for clinical diagnosis.
  • Existing methods often lack the simplicity, affordability, or field-suitability required for widespread use.
  • There is a persistent need for cost-effective and robust hemoglobin assays for field conditions.

Purpose of the Study:

  • To evaluate a novel cyanmethaemoglobin-based hemoglobin assay utilizing microtitre plates.
  • To assess the feasibility and performance of this method for rapid hemoglobin estimation in field settings.

Main Methods:

  • A cyanmethaemoglobin-based assay was developed on microtitre plates.
  • The new method was validated against a standard haematology analyser using samples from voluntary blood donors.
  • Assay stability, reproducibility, linearity, and cost-effectiveness were rigorously evaluated.

Main Results:

  • Optimal results were achieved with 10 μL blood in 300 μL Drabkin's solution after 10 minutes incubation.
  • Color stability extended up to 6 hours, with a coefficient of variation below 3%.
  • The assay demonstrated a low cost (<3 cents/test) and rapid turnaround time (30 minutes for 90 samples).

Conclusions:

  • Microtitre plate-based cyanmethaemoglobin assay is a feasible, robust, and rapid method.
  • The technique is cost-effective and suitable for hemoglobin estimation in field conditions.
  • This assay addresses the need for simple, field-deployable diagnostic tools.
