Overview Of Cell Separation And Isolation

7.7K
Cell separation was first achieved in 1964 by S. H. Seal, who separated large tumor cells from the smaller blood cells using filtration. Two years later, Pohl and Hawk performed experiments on how cells respond differently to a nonuniform electric field based on the cell type. Such observations were the inception of cell separation methods, which allow isolating a single cell type from a heterogeneous sample.

Summary
This summary is machine-generated.

Isolate® is a suitable alternative to Percoll® for sperm selection in bovine assisted reproductive techniques. Both methods yield similar sperm quality and embryo development, offering a viable option for in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI).

Area of Science:

  • Veterinary Science
  • Reproductive Biology
  • Biotechnology

Background:

  • Sperm selection is crucial for assisted reproductive techniques (ART) like in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI).
  • Percoll® is a widely used medium for sperm selection, but alternatives are sought for ART applications.

Purpose of the Study:

  • To evaluate Isolate® as a potential substitute for Percoll® in selecting bovine spermatozoa for ART.
  • To compare the quality of spermatozoa selected using Isolate® versus Percoll®.

Main Methods:

  • Commercial cryopreserved bovine semen was subjected to sperm selection using both Isolate® and Percoll® gradients.
  • Sperm quality parameters including motility (CASA), plasma and acrosome membrane integrity, reactive oxygen species (ROS) levels, DNA integrity, and mitochondrial membrane potential (ΔΨm) were assessed by flow cytometry.
  • In vitro embryo production was evaluated for all experimental groups.

Main Results:

  • Both Isolate® and Percoll® demonstrated similar percentages of spermatozoa with intact plasma membrane, acrosome integrity, and high mitochondrial membrane potential (ΔΨm).
  • Percoll® showed a higher percentage of spermatozoa with intact plasma and acrosome membrane compared to the post-thawing group.
  • No significant differences were observed in motility, ROS levels, DNA fragmentation, or in vitro embryo production between the Isolate® and Percoll® groups.

Conclusions:

  • Isolate® provides comparable sperm quality and embryonic development outcomes to Percoll® for bovine ART.
  • Isolate® serves as a suitable alternative sperm selection method for bovine assisted reproductive techniques, offering similar efficacy to Percoll®.
